Philadelphia (Ph) chromosome, formed via translocation t (9; 22), is the cytogenetic hallmark of CML. The translocation forms a chimeric gene, bcr-abl, that generates BCR-ABL, a fusion protein that constitutively activates ABL tyrosine kinase and causes CML.
